500 MHz TCI Cryoprobe Spectrometer - 'Glenlivet' - Room B19

One of our two 500MHz instruments in B19, the Avance 500 TCI Cryoprobe system enhances our abilities in the area of protein chemistry.  With its Triple Resonance Inverse (TCI) Automatic Tuning and Matching (ATM) cryoprobe, CHN combination experiments are possible.  Prof. Chris Dobson's research group make use of this instrument, together with our highest field 700 MHz spectrometer for these types of studies.  However, the suitability for experiments such as 3D TOCSY-NOESY, WATERGATE presaturation and other techniques together with a 60 position sample changer, mean that this instrument is also in demand from those groups who are conducting research with smaller molecules, and those using water as a primary solvent.

This instrument is equipped with a 5mm TCI ATM cryoprobe with Z-Gradients.  TCI probes are specially designed to allow a reasonable sensitivity to carbon nuclei as well as nitrogen and proton, whereas the older TXI style probe had very poor carbon performance.  We still advise however that carbon experiments on samples of low concentration should be carried out on the Service DCH 500 cryoprobe.

The instrument is run by members of the NMR Service as required, and also by trained group users.  A wide variety of experiments are available for use including 1H, DQF-COSY, HMQC, HSQC, HMBC, TOCSY, ROESY and NOESY, together with Gradient nOe, WATERGATE 1H and presaturation versions of 2D experiments.  There are also parameter sets for the major groups of experiments used by our protein chemists - we will be able to advise on the best people to talk to if you require more help in this area, and can arrange for Bruker specialists to visit if something very complex requires setting up.  Most users will find the basic library of programs cover their needs with minor adjustments to suit.  Hard and soft, as well as decoupler pulse calibration is needed to get the most out of certain samples however.  Please talk to the NMR service if you need assistance - we can usually help and get your experiment running quickly and safely.
